With the death of her father, Eun-chan has to support her family, consisting of a foolish, spendthrift mother and younger sister. Due to her short hair, baggy clothing and general boyish behavior, she often gets mistaken for a boy and her sister often uses her as her 'boyfriend' to get rid of boys who follow her around. Eun-chan is also proficient at Taekwondo, and is strong for a girl.

Han-kyul is the heir of a large company. He's rich, carefree and doesn't take life seriously enough. He first meets Eun-chan when one of her sister's boyfriends attempts to steal a handbag belonging to Han Yoo-ju (a woman that Han-kyul secretly likes for nine years). Eun-chan, not knowing who the boy is, catches him but lets him go when she realizes who it is. This gives Han-kyul the impression that the two have been planning the incident together, and when she comes the day after to collect money to fix her scooter, he insults her by telling her that he thinks she's just a thief. All this time, he thinks that Eun-chan is a male.

Meanwhile, Han-kyul's cousin, Choi Han-seong, is an established music producer and his ex-girlfriend Han Yoo-ju, is a famous artist who studied in New York. The two were lovers for about 8 years but their relationship ended when Yoo-ju broke up with Han-seong for another man. Two years after they separated their paths cross again when Yoo-ju returns to Seoul to make up with Han-seong, who was smitten by Eun-chan's "yup-gi" (disgusting but lovable) personality. He knows Eun-chan is a girl for she delivers milk in his house, and agrees to keep her gender secret from everyone.

After being forced by his grandmother to take on a coffee shop (that is to become Coffee Prince) and improve its sales, Han-kyul goes for a 'Prince' concept and tries to hire good-looking boys as servers to attract female customers. Eun-chan is employed. Eun-chan at first liked Han-seong, but her feelings for him vanish as she works at Coffee Prince, she found herself in love with Han-kyul. Han-kyul too feels affection for Eun-chan, but not knowing that Eun-chan is actually a girl, this leads him to confusion about his sexuality and he tries to distance himself from her.

Eun-chan is definitely a manly-looking girl. Since she was in high school, girls of her age had often mistaken her for a pretty boy and tried to date her. Since her father passed away, she has worked at all kinds of jobs to support her family. Despite her difficult situation, she maintains a courageous and forthright demeanor. A likeable person, she enjoys eating good food and is friendly to everyone. Influenced by her coffee-loving father, she has an uncanny ability to judge the aroma of coffee. Thanks to this ability, she lands a job at Coffee Prince and dreams of becoming a barista one day. She falls in love with Han-kyul who is cold and wild, and since their encounter, her life is no longer as peaceful as it had been.
 

Gong Yoo as Choi Han-kyul
Han-kyul is the heir to the Dongin Foods Company. He had been working in the U.S., but returns to Korea at his grandmother’s request. Smart, witty, and handsome, he is popular with women. He hasn’t settled on any particular job, and still has so many things he wants to do. Clearly, taking over the family business is not what he has in mind. He is unable to defy his grandmother’s requests, however, so he starts managing the coffee shop, Coffee Prince, and grows to enjoy the taste of its coffee. Definitely stubborn, he doesn’t want to ask for favors or give them. Likewise, he acts with indifference toward others. However, deep inside he is warm hearted. He meets Eun-chan, a lively boy (who is actually a girl) and falls in love with him, which has him deeply troubled about his sexual preference.
 

Lee Seon-gyeon as Choi Han-seong
Not only does Han-seong have a sweet voice and smile but he is also easy-going, so he can easily make friends with anyone, but he doesn’t reveal his true feelings. Like other artists, his is stubborn and somewhat eccentric. A musician working at a broadcast company, he lives alone - away from his family and the family business, Dongin Foods. As far as work is concerned, he is full of confidence and active. He has found it difficult to be close to women ever since he broke up with his ex-girlfriend, Yoo-ju. However, he finds he is drawn to Eun-chan who befriends him.
 

Chae Jung-an as Han Yoo-ju
Yoo-ju is a stylish and sexy artist. The raven-haired beauty has been working actively in both New York and Seoul. She is liberal to the point of selfishness, wearing whatever fashion suits her taste. She makes friends with others quite easily but her unrestrained character often leads to loneliness. Deep inside, she is hurt easily and has trouble getting over it. She once loved Choi Han-seong but her passion for art causes her to break up with him and go to New York. She later goes back to Han-seong only to learn the painful truth - that he doesn’t feel the same way about her
